Figure S1. High Ca2+i shifts the gating towards th e fully liganded states for Scheme VII. Equilibrium occupancies of the indicated open and closed states in Scheme VII are plotted on a log scale at low (A) and high (B) Ca2+i. The x axis indicates the number of bound Ca2+ , and the y axis indicates the rows of three closed and two open states. The inset identifies the states in the plots with the state numbers in Scheme VII. At 5.5 µM Ca2+i, Scheme VII predicts that the channel wou ld spend most of its time in the two closed states with zero or one bound Ca2+. At 1,024 µM Ca2+i, Scheme VII predicts that the channel would spend nearly all of its time in the three closed and two open stat es with four bound Ca2+. Channel B06.

Figure S2. Estimated rate constants for Schemes IV-VII, XII, and XIII for three BK channels: B06 (black bar), B12 (hatched bar), and B14 (light gray bar). Rate constants in A-D were estimated by the simultaneous fitting of 2-D dwell-time distributions at six different Ca2+i. Rate constants in E-F were estimated by the fitting of the 2-D dwell-time distributions obtained at 1,024 µM Ca2+i only.
